Título Susceptibilidade da glândula mandibular de lagartas híbridas de “Bombyx mori” (Lepidoptera: Bombycidae) ao nucleopolyhedrovirus múltiplo

Resumo O Bombyx mori nucleopolyhedrovirus tipo múltiplo (BmMNPV) foi utilizado na inoculação de lagartas híbridas de Bombyx mori do 5º ínstar para analisar a susceptibilidade das células da glândula mandibular. Esta glândula é de fundamental importância para vida do inseto, uma vez que assume o papel da glândula sericígena na produção e secreção de saliva, indispensável no processo de alimentação. As glândulas foram coletadas e analisadas em diferentes tempos pós-inoculação (dpi), assim como o corpo gorduroso e traquéias, que foram utilizados como controle positivo da infecção pelo BmMNPV. Os resultados evidenciaram que as células da glândula mandibular não são susceptíveis ao BmMNPV em nenhum dos tempos analisados. No entanto, as células do corpo gorduroso e traquéias revelaram aspectos característicos de infecção viral a partir do 5º dpi, confirmando a virulência do inóculo. A análise histológica da glândula mandibular revelou traquéias somente na periferia da glândula mandibular, não no interior no tecido. A detecção de polissacarídeos neutros (PAS) evidenciou a presença da lâmina basal (LB) intacta e delimitada, na superfície da membrana em contato com a hemolinfa, envolvendo a glândula mandibular. A ausência de infecção das células da glândula mandibular pode ser decorrente de vários fatores como: (1) a presença da LB, atuando como uma barreira seletiva; (2) a não penetração do sistema traqueal, uma vez que este órgão é considerado um importante dispersor da infecção nos insetos; (3) expressão gênica diferencial do genoma viral, com o objetivo de prolongar a sobrevida do hospedeiro (enzimas secretadas pela glândula mandibular auxiliam na alimentação da larva), permitindo desse modo, a disseminação do BmMNPV em vários tecidos larvais.

Abstract The Bombyx mori nucleopolyhedrovirus, subtype multiple, BmMNPV, was used for inoculation of 5th instar Bombyx mori hybrid larva to analyze the mandibular gland cells baculovirus susceptibility. This gland has a fundamental importance in insect’s life as it assumes the role of the silk gland in the production and secretion of saliva, which is essential in the feeding process. To perform that, the glands were collected and analyzed at different time’s post-inoculation (dpi), as well as, the fat body and trachea, as positive control to BmMNPV infection. The results showed that the mandibular gland cells are not susceptible to BmMNPV in any of the times analyzed, however, the cells in the fat body and trachea revealed characteristic features of viral infection from 5 dpi, which confirms the virulence of the inoculum. Histological analysis revealed the presence of the tracheae only at the periphery of the gland, not into the tissue. The detection of neutral polysaccharides (PAS) revealed the presence of basal lamina (LB) intact and bounded the membrane surface in contact with the hemolymph. The absence of infection of cells in the mandibular gland may be due to several factors: (1) the presence of LB, acting as a selective barrier; (2) no penetration of the tracheal system, since this organ is considered an important to insect’s infection spread; (3) differential gene expression of the viral genome in order to prolong the survival of the host (the mandibular gland secreted enzymes that assist the larva feeding), thereby allowing the BmMNPV infection in several larval tissues.
