Resumo A ferrugem asiática da soja (FAS), causada pelo fungo Phakopsora pachyrhizi, é uma doença foliar destrutiva em quase todos os países produtores de soja. Ferramentas biotecnológicas podem auxiliar no entendimento dos mecanismos de resposta de defesa a este fungo em nível molecular e consequentemente no controle da doença. Assim, o objetivo deste trabalho foi identificar e analisar a expressão diferencial de genes e o acúmulo de compostos fenólicos para obter maiores informações a respeito da resposta da soja à infecção. Para a identificação de genes diferencialmente expressos (DEGs), RNA de folhas infectadas e não infectadas de genótipos de soja resistente (PI561356) e suscetível (BRS 184) foram analisadas pela metodologia ACP (Annealing Control Primer). Foram obtidos 44 DEGs com homologia a proteínas conhecidas, envolvidas principalmente em fotossíntese, síntese e degradação de proteínas. Para as análises em RT-qPCR e em cromatografia líquida de alta performace (HPLC), foram utilizados os mesmos genótipos resistentes e suscetíveis e foi adicionado o genótipo tolerante (BRS 231). A maioria dos genes analisados apresentaram perfis de expressão semelhantes entre os genótipos avaliados, sendo mais ativados em 192 horas após a inoculação (hai), demonstrando um envolvimento com a resposta basal de defesa. No entanto, alguns genes foram ativados mais precocemente e intensamente nas plantas resistentes, sugerindo um maior envolvimento com a resistência. As análises de HPLC demostraram um acúmulo das isoflavonas daidzeína e genisteína nos três genótipos avaliados em resposta à infecção, principalmente nos horários de 192 hai, sugerindo uma resposta basal ao ataque do fungo. No entanto, o acúmulo mais intenso de isoflavona na forma livre aglicona, em horários mais iniciais, na PI561356, pode estar envolvido com a resistência dessas plantas. Adicionalmente, foi possível identificar genes candidatos para estudos mais detalhados de seu envolvimento com a resistência à ferrugem asiática da soja.

Abstract Asian soybean rust (ASR), caused by Phakopsora pachyrhizi, is a destructive foliar disease in nearly all soybean-producing countries. Biotechnology tools may help to understand the plant host response to this fungus at molecular level and subsequently for control of the disease. The objective of this study was to identify and analyze the differential gene expression and accumulation of phenolic compounds for more information about the soybean response to infection. To identify differential expression genes (DEGs), RNA from rust-infected and non-infected leaves of resistant (PI561356) and susceptible (BRS 184) genotypes of soybean were analyzed by the Annealing Control Primer (ACP) methodology. Forty four DEGs were obtained with homology to know proteins, mainly involved in photosynthesis, protein synthesis and degradation. For the RT-qPCR and high performance liquid chromatography (HPLC) analyzes were used the same resistant and susceptible genotypes and was adiconado the tolerant genotype (BRS 231). For most of the genes analyzed expression profiles were similar among genotypes and activation was higher at 192 hours after inoculation (hai) what demonstrate their involvement in the basal defense response. On the other hand, some genes were intensely activated earlier in the resistant plants suggesting a greater involvement in the resistance. HPLC analysis showed accumulation of the isoflavones genistein and daidzein in all genotypes in response to the infection, particularly at 192 hai, suggesting a basal response to the fungal attack. Early intense accumulation of free aglycone in the resistant genotype (PI561356) may be involved in the resistance of these plants. Additionally, it was possible to identify candidate genes for further studies on its involvement in the resistance to Asian soybean rust.
