Título RNA mensageiro das proteínas HSP70 e MJP3 em abelhas Apis mellifera L. (Hymenoptera:Apidae) após suplementação alimentar com proteínas, prebióticos e probióticos

Resumo Abelhas da espécie Apis mellifera são importantes como polinizadoras, ao compararmos a outros insetos. Isso acontece pelo seu grande número na natureza e pela facilidade de adaptação as mais diversas estruturas florais, aumentando de forma significativa o rendimento de sementes e frutos, sendo em algumas regiões, responsáveis por até 90% da polinização local. Dentre os produtos processados pelas abelhas estão o mel, cera, própolis, pólen apícola, geleia real e apitoxina. A geleia real (Royal Jelly - RJ) das abelhas é estudada quanto a sua composição em substâncias fisiologicamente ativas, por propiciar a diferenciação de uma larvas com a mesma composição genética, em uma rainha ou operária. As Proteínas Principais da Geleia Real – MRJPs são um grupo de proteínas que têm origem evolutiva comum com proteínas Yellow de Drosophila melanogaster, e são expressas pelas glândulas hipofaringeanas de abelhas enfermeiras, a MRJP3 é uma dessas proteínas. Pesquisas mostraram que existe uma alta correlação entre a produção de MRJP3 e a produtividade da geleia real. Já as Proteínas de Choque Térmico (HSPs), são responsáveis pelas respostas metabólicas do organismo quando exposto a algum tipo de estresse, e a família HSP70 é uma das que mantém suas sequências mais conservadas, sendo importante no dobramento de proteínas, por isso, é uma das mais estudadas quanto à resposta celular. O objetivo deste estudo foi suplementar a alimentação de colônias de abelhas A. mellifera, com alimentos prebióticos e probióticos e, posteriormente avaliar por meio de qRT-PCR os níveis expressão das proteínas MRJP3 e HSP70. Assim, abelhas recém emergidas foram mantidas em gaiolas experimentais por um período de 11 dias. Foram realizados quatro tratamentos: ração proteica base, ração proteica prebiótica e probiótica, controle negativo contendo apenas pólen, xarope de açúcar e água, e controle positivo que continha os dois tipos de ração. Durante o experimento foram realizadas três coletas. Pelo método de reação em cadeia da polimerase em tempo real (qRT-PCR), foi possível analisar que, valores médios de 2-.CT dos transcritos de RNA mensageiro do gene MRJP3 e HSP70, apresentaram alterações na expressão de mRNAs de acordo com o dia de coleta e tratamento submetido. Com a técnica de qRTPCR, foi possível observar que os níveis médios de transcritos de mRNA são maiores no quarto dia de tratamento para MRJP3, diminuindo no décimo primeiro dia, período que também é registrado uma diminuição das atividades das glândulas hipofaringeanas. Os mRNAs de HSP70 apresentaram aumento de expressão desde o quarto dia até o final do tratamento com prebiótico e probiótico, provavelmente devido a modificações do metabolismo intermediário provocado por tais agentes. Finalmente, podemos considerar que a ração protéica contendo prebióticos e probióticos é um suplemento alimentar que promove alterações na expressão gênica de A. mellifera.

Abstract Bees of the species Apis mellifera are important as pollinators when compared to other insects. This is due to its large number in nature and the ease of adaptation to the most diverse floral structures, significantly increasing the yield of seeds and fruits, being in some regions, responsible for up to 90% of local pollination. Among the products processed by bees are honey, wax, propolis, bee pollen, royal jelly and apitoxin. The royal jelly (Royal Jelly - RJ) of the bees is studied for its composition in physiologically active substances, for allowing the differentiation of a larvae with the same genetic composition, in a queen or worker. The Principal Proteins of Royal Jelly - MRJPs are a group of proteins that have common evolutionary origin with Yellow proteins of Drosophila melanogaster, and are expressed by the hypopharyngeal glands of bees nurses, MRJP3 is one of these proteins. Research has shown that there is a high correlation between the production of MRJP3 and the productivity of the royal jelly. On the other hand, the HSPs are responsible for the metabolic responses of the body when exposed to some type of stress, and the HSP70 family is one of the most conserved sequences, being important in protein folding. one of the most studied as regards the cellular response. The objective of this study was to supplement the feeding of colonies of A. mellifera bees with prebiotic and probiotic foods and later to evaluate the expression levels of MRJP3 and HSP70 proteins by means of qRT-PCR. Thus, newly emerged bees were kept in experimental cages for a period of 11 days. Four treatments were performed: base protein ration, prebiotic and probiotic protein ration, negative control containing only pollen, sugar syrup and water and positive control containing the two types of feed. During the experiment three collections were carried out. By means of the real-time polymerase chain reaction (qRTPCR) method, it was possible to analyze that mean values of 2-.CT of MRJP3 and HSP70 messenger RNA transcripts showed alterations in mRNA expression according to the day of collection and treatment submitted. With the qRT-PCR technique, it was possible to observe that the mean levels of mRNA transcripts are higher in the fourth day of treatment for MRJP3, decreasing on the eleventh day, a period also showing a decrease in the activities of the hypopharyngeal glands. HSP70 mRNAs increased expression from the fourth day to the end of prebiotic and probiotic treatment, probably due to modifications of the intermediate metabolism caused by such agents Finally, we can consider that the protein diet containing prebiotics and probiotics is a food supplement that promotes changes in the gene expression of A. mellifera.
