Título O impacto de retrotransposons na indução de polimorfismo genético e na geração de cultivares de uvas de mesa (Vitis vinifera L.)

Resumo A cultivar (cv.) Itália (Vitis vinifera) foi desenvolvida no ano de 1911 na Itália, e no Brasil foi introduzida na década de 20. Em Marialva, Noroeste do estado do Paraná, no ano de 1962 iniciou-se a produção desta cultivar, e assim, este município ficou conhecido como a Capital da Uva Fina de Mesa. As ocorrências de mutações somáticas deram origem as uvas de cor. Mutações somáticas em Itália deram origem as cultivares Rubi e Benitaka. Mutações em brotações nos parreirais de Benitaka originaram a cv. Brasil e mutações na cv. Brasil deram origem a cv. Black Star. Elementos transponíveis são descritos como os maiores contribuintes para promover mutações somáticas, e grande parte do genoma das plantas é constituído por estas sequências. Os Elementos transponíveis apresentam um importante papel na domesticação e no melhoramento genético da videira. A inserção do retrotransposon Gret1 na região promotora do gene VvmybA1, e uma mutação de um único nucleotídeo (SNP) no gene VvmybA2 levam a inibição da expressão do gene UFGT que é responsável pela produção de antocianina nas bagas das uvas de cor, deste modo, a cv. Itália possui a baga branca. E para as uvas de cor, Rubi, Benitaka, Brasil e Black Star ocorreu a restauração do gene VvmybA1, ativando assim a via da biossíntese de antocianina. No presente estudo foram avaliadas amostras de DNA das cinco cultivares de uvas finas de mesa, Itália, Rubi, Benitaka, Brasil e Black Star, coletadas em uma propriedade do município de Marialva, Paraná. O objetivo geral deste trabalho foi entender a participação de retrotransposons no desenvolvimento destas cinco cultivares de uva. No capítulo um foi abordado o desenvolvimento dos primers para as sequencias LTR. Estes foram criados a partir de informações de bancos de dados do genoma de Vitis. Com o uso destes primers foram realizadas análises com marcadores IRAP (Interretrotransposon Amplified Polimorphism) e REMAP (Retrotransposon- Microssatellite Amplified Polymorphism). O polimorfismo obtido com os dois tipos de marcadores foi de 49%, e foi constado que a maior variação genética encontra-se dentro de cada uma das cinco cultivares. Foi identificado que cerca de 72% das plantas da cultivar Black Star agrupou-se com a cv. Benitaka. No capítulo dois foi abordado o uso das informações da literatura sobre o retrotransposon Gret1 e o gene VvmybA1, para amplificar e estudar esses segmentos nas cinco cultivares. Seis combinações de primers foram utilizadas, e três alelos foram encontrados, VvmybA1(ITA), VvmybA1(RUO) e VvmybA1(BEM). A cultivar Itália é homozigota para este loco (ITA/ITA), as cultivares Brasil, Benitaka e Black Star são heterozigotas (ITA/BEN) e a cultivar Rubi é heterozigota (ITA/RUO). A partir do sequenciamento dos produtos amplificados observou-se uma mutação de nucleotídeo único (SNP) no éxon 1. A trinca de nucleotídeos (ATC) que codifica o aminoácido isoleucina nas cultivares Rubi e Itália foi substituído pela trinca (ACC) que codifica o aminoácido treonina em Benitaka, Brasil e Black Star. Foram observadas alterações no éxon 3, mas estas não puderam ser relacionadas com alterações na sequência de aminoácidos. Alterações relacionadas com mutações de ponto e In/Del foram observadas nos introns 1 e 2. Estas alterações podem estar associadas com diferenças na taxa de processamento de RNAm do gene VvmybA1 interferindo na coloração das bagas.

Abstract The cultivar Itália (Vitis vinifera) was developed in 1911 in Italy, and the cultivar Brazil it was introduced in the 1920’s. In Marialva, Northwest of the state of Paraná, in 1962 started the production of this cultivar, and Marialva is known as the Capital of Table Grapes. The occurrence of somatic mutations gave rise to grapes of color. Somatic mutations in Itália originated the cultivars Rubi and Benitaka. Mutations in shoots in the Benitaka vineyards originated cv. Brazil and mutations in cv. Brazil gave rise to cv. Black Star. Transposable elements are described as the major contributors to promoting somatic mutations, and much of the plant genome consists of these sequences. The Transposable Elements play an important role in the domestication and genetic improvement of the vine. The insertion of the Gret1 retrotransposon into the promoter region of the VvmybA1 gene, and a single nucleotide mutation (SNP) in the VvmybA2 gene leads to inhibition of the expression of the UFGT, the gene that is responsible for the production of anthocyanin in the berries of color grapes, thus cv. Itália has the white berry. For the grapes of color, Rubi, Benitaka, Brazil and Black Star, the restoration of the gene VvmybA1 occurred, thus activating the anthocyanin biosynthesis pathway. In the present study, DNA samples of the five cultivars of table grapes, Itália, Rubi, Benitaka, Brazil and Black Star, collected from a property of the municipality of Marialva, Paraná, were evaluated. The general objective of this work was to understand the participation of retrotransposons in the development of these five grape cultivars. In Chapter One the development of the primers for the LTR sequences was discussed. These were created from information from databases of the genome of Vitis. These primers were analyzed with IRAP (Interretrotransposon Amplified Polimorphism) and REMAP (Retrotransposon- Microsatellite Amplified Polymorphism) markers. The polymorphism obtained with the two types of markers was 49%, and it was recorded that the greatest genetic variation is found within each of the five cultivars. It was identified that about 72% of the plants of the cultivar Black Star were grouped with cv. Benitaka. In Chapter Two, the literature information on the Gret1 retrotransposon and the VvmybA1 gene was discussed, to amplify and study these segments in the five cultivars. Six primer combinations were used, and three alleles were amplified, VvmybA1(ITA), VvmybA1(RUO) and VvmybA1(BEN). The cultivar Itália is homozygous for this locus (ITA/ITA), the cultivars Brazil, Benitaka and Black Star are heterozygous (ITA/BEN) and the cultivar Rubi is heterozygous (ITA/RUO). From the sequencing of the amplified products, a single nucleotide mutation (SNP) was observed in exon 1. The codon (ATC) encoding the amino acid isoleucine in the Rubi and Itália cultivars was replaced by the codon (ACC) encoding the threonine amino acid in Benitaka, Brazil and Black Star. Changes were observed in exon 3, but these could not be related to changes in the amino acid sequence. Changes related to point mutations and In/Del were observed in introns 1 and 2. These changes may be associated with differences in the processing rate of mRNA of the VvmybA1 gene interfering with the coloring of the berries.
