Resumo A antracnose, causada pelo fungo Colletotrichum lindemuthianum, é uma das principais doenças que incidem na cultura do feijão comum. Esse fungo possui ampla diversidade de virulência, caracterizada por raças Mesoamericanas e Andinas. Genes de resistência à antracnose de origem Andina frequentemente conferem resistência às raças Mesoamericanas, as quais são consideradas mais virulentas. Os objetivos deste trabalho foram estudar a resistência genética à antracnose na cultivar Andina de feijão comum Beija Flor; mapear o locus de resistência à antracnose; e identificar marcadores moleculares ligados a esse locus. Foram utilizados dois cruzamentos, Beija Flor × Cornell 49242, cujo as famílias F2:3 foram inoculadas com a raça 1545 e Beija Flor × Crioulo 159, em que as plantas F2 foram inoculadas com as raças 4, 321, 453 e 1545 de C. lindemuthianum. Foi utilizado análise de bulk segregante para genotipagem das plantas F2 dos dois cruzamentos. O DNA dos bulks e dos parentais foram analisados com 11.292 marcadores SNPs utilizando o Illumina BeadChip BARCBean12K. Marcadores do tipo Kompetitive Allele Specific PCR (KASP) foram utilizados para o fine mapping do locus de resistência à antracnose da cultivar Beija Flor. Os resultados dos testes de herança indicaram que Beija Flor apresenta um único gene de efeito dominante, o qual confere resistência as raças 4, 321, 453 e 1545 de C. lindemuthianum. O locus de resistência à antracnose foi posicionado no cromossomo Pv04, entre os marcadores SS333 (422,293 pb) e SS309 (454,032 pb), espaçando uma região de 31,7 kpb, nessa região se encontram três genes candidatos, que podem conferir resistência à antracnose na cultivar Andina Beija Flor.

Abstract Anthracnose, caused by the Colletotrichum lindemuthianum fungus, is one of the main diseases affecting the common bean crop. This fungus has a wide diversity of virulence, characterized by Mesoamerican and Andean races. Anthracnose resistance genes of Andean origin often confer resistance to Mesoamerican races, which are considered more virulent. The objectives of this work were to study the genetic resistance to anthracnose in the Andean common bean cultivar Beija Flor; map the anthracnose resistance locus; and identify molecular markers linked to that locus. Two crosses were used, Beija Flor × Cornell 49242, which F2:3 families were inoculated with race 1545 and Beija Flor × Crioulo 159, which the F2 plants were inoculated with races 4, 321, 453 and 1545 of C. lindemuthianum. Bulk Segregating analysis was performed for genotyping the F2 plants from the both crosses. DNA from the bulks and parental were analyzed with 11,292 SNPs markers using the Illumina BeadChip BARCBean12K. Kompetitive Allele Specific PCR (KASP) markers were used for the fine mapping of the anthracnose resistance locus in the Beija Flor. The results of the inheritance tests indicated that Beija Flor has a single dominant gene, which confers resistance to races 4, 321, 453 and 1545 of C. lindemuthianum. The anthracnose resistance locus was positioned on chromosome Pv04, between the SS333 (422,293 bp) and SS309 (454,032 bp) markers, spacing a region of 31.7 kbp, in this region there are three candidate genes that could confer resistance to the anthracnose pathogen in the Andina cultivar Beija Flor.
