Resumo |
Os estresses bióticos e abióticos são fatores que resultam na redução da produtividade de diversas culturas anualmente. O melhoramento genético de plantas, aliado à biotecnologia, tem possibilitado o desenvolvimento de genótipos mais tolerantes a esses fatores. Assim, a transgenia em plantas tem sido empregada para a introdução de genes que desempenham papel de osmoprotetores, a fim de obter cultivares tolerantes aos estresses abióticos. A prolina é considerada um destes osmoprotetores, logo, a enzima Δ¹-pirrolina-5-carboxilato sintetase (P5CS), codificada pelo gene p5cs, é fundamental e limitante na rota de síntese de prolina. Portanto, este trabalho teve como objetivo avaliar o acúmulo de prolina em plantas de trigo transformadas com o gene o p5cs. Os resultados obtidos indicaram que as plantas transgênicas e as plantas não irrigadas (controle) sofreram redução no acúmulo de água em relação às plantas irrigadas (controle), porém suportaram oito dias sem o suprimento de água e com turgescência. Por outro lado, em relação ao acúmulo de prolina, as plantas transgênicas produziram aproximadamente 2,1 vezes mais do que as plantas controle submetidas ao estresse hídrico, demonstrando que houve a expressão do gene p5cs. Os teores de prolina nas plantas transgênicas com a construção promotor constitutivo (Ubi) e no promotor estresse induzido (aipc) não apresentaram diferença estatística significativa. Os valores de peroxidação de lipídeos não diferiram estatisticamente entre as plantas transgênicas e as plantas controle com estresse hídrico no período de avaliação. Este fato sugere que apesar dos elevados teores no acúmulo de prolina entre as plantas transgênicas, não houve a formação de mecanismos eficientes de proteção nas membranas celulares contra o estresse oxidativo. |

Abstract |
The biotic and abiotic stresses are factors that result in yield reduction in several crops annually. The plant breeding, associated with biotechnology, has allowed the development of genotypes more tolerant to these factors. Thus, transgenic process in plants has been used to introduce genes that perform as osmoprotector, in order to obtain tolerant cultivars to abiotic stresses. The proline is considered one of these osmoprotectors, therefore, the enzyme Δ¹-pirrolyne-5-carboxylate synthetase (P5CS), codificated by p5cs gene is important and limiting in the synthesis pathway of it. Hence, this work had as objective to evaluate the accumulation of proline in wheat plants transformed with p5cs gene. The results obtained indicated that transgenic plants and not irrigated plants (control) had suffered reduction of water accumulation in relation to plants irrigated (control), but they bore eight days without water and with turgescence. On the other hand, in relation to proline accumulation, transgenic plants produced approximately 2.1 more than control plants, demonstrating that there was an expression of p5cs gene. The transgenic plants with constituted promoter (Ubi) and induced stress promoter (aipc) had not presented significant differences in relation to proline levels, being the events statistically equal. The lipids peroxidation values did not show statistic differences between the transgenic plants and the not irrigated control plants in the evaluation period. This fact suggests that despite of high levels in accumulation of proline between transgenic plants, there was no production of efficient mechanisms to protect cellular membranes against oxidative stress. |
