Resumo O lepidóptero Bombyx mori L. é um inseto totalmente domesticado para a utilização comercial e é continuamente selecionado geneticamente para otimizar qualitativa e quantitativamente a produção de seda. A atividade sericícola, que compreende a criação de lagartas de B. mori e a venda dos casulos para as indústrias de fiação, tem como uma das principais causas de perda na produção a ocorrência de doenças causadas por vírus, como os Nucleopolyhedrovirus (NPV). O NPV pode conter apenas um nucleocapsídeo por vírion (Singie Nuclear Polyhedrosis Virus - SNPV) ou vários nucleocapsídeos por vírion (Múltiplo Nuclear Polyhedrosis Virus - MNPV). Com o objetivo de verificar diferenças genéticas no isolado geográfico do Paraná, o Bombyx mori nucleopolyhedrovirus subgrupo múltiplo. BmMNPV, em relação ao BmNPV denominado singie e de ocorrência geral, foram amplificadas e clonadas as ORFs 10 e 14 do BmMNPV. As análises in silico indicaram que a ORF 10 apresenta 70 aminoácidos e alta identidade com alguns isolados geográficos de BmNPV. A análise da sequência de aminoácidos da ORF 14 apresentou alterações na sequência e no perfil de hidrofobicidade em relação à ORF 14 do BmNPV. Adicionalmente a análise in silico dessa proteína indica que, provavelmente, a ORF 14 se localiza no envelope virai e está associada à resposta ao stress, como a causada pela infecção do baculovirus nas células alvo de Bombyx mori.

Title Anafysis of ORF 10 and 14 of the Bombyx mori, nucleopolyhedrovirus multiple subgroup, BmMNPV
Abstract The Bombyx mori L. is an insect entirely domesticated for the commercial use and is continuosly selected genetically to optimize qualitatively and quantitatively the silk production. The activity of the sericulture, that includes the rearing of B. mori caterpillars and the cocoons production for the wiring's industries, has as the one of the main causes of production loss the occurrence of the diseases caused for virus, as Nucleopolyhedrovirus (NPV). The NPV contain only one nucleocapside for virion (Nuclear Single Polyhedrosis Virus - SNPV) or several nucleocapsides for virion (Nuclear Multiple Polyhedrosis Virus - MNPV). With the objective to analysis genetic differences in the Bombyx mori nucleopolyhedrovirus multiple subgroup, BmMNPV, a geographic isolated from Paraná, in relation to the called BmNPV single and of general occurrence, were amplified, cloned and performed the in silico analysis of the BmMNPV ORFs 10 and 14. The in sílico studies indicated that ORF 10 presents 70 amino acids and higher identity with some BmNPV geographic isolates. The BmMNPV ORF 14 amino acid sequence analysis showed alterations in the sequence and the hydrophobicity profile in relation to BmNPV ORF 14. Additionally the ín silico analysis of this protein indicates that, probably, ORF14 is located in the viral envelope and is associated with the stress response, as caused for the baculovirus infection in the Bombyx mori cells.
